Why Are Some Tissue Culture Media Black? Activated Charcoal and Banana in Plant Tissue Culture
During a recent visit to a plant tissue culture facility, we noticed something interesting.
The plantlets themselves did not appear unusual.
What immediately caught our attention was the culture medium at the bottom of the vessels.
It was very dark—almost black.
Our first thought was:
Does the medium contain activated charcoal?
After speaking with the team at the facility, we learned that activated charcoal was indeed used in this culture system, together with banana as an organic additive.
For professionals working with orchid tissue culture, this may not be particularly surprising.
Activated charcoal has been used across a range of plant tissue culture applications, while banana homogenate and other natural organic supplements can also be found in some orchid culture protocols.
So why are these ingredients added to tissue culture media?
1. Why Are Some Plant Tissue Culture Media Black?
When a tissue culture medium appears very dark or almost black, activated charcoal is one of the first possible causes to consider.
Adding activated charcoal significantly darkens the medium.
However, an important distinction should be made:
Darkening the medium is an effect of activated charcoal, not the primary reason for using it.
The more important property of activated charcoal is its:
high adsorption capacity.
Activated charcoal has an extensive porous structure and large internal surface area, allowing it to adsorb a variety of substances from the culture medium.
This property explains much of its long-standing use in plant tissue culture.
2. What Does Activated Charcoal Do in Plant Tissue Culture?
Plant tissues maintained under in vitro conditions may release phenolic compounds and other metabolites into the culture environment.
In certain culture systems, the accumulation of these substances can negatively affect the cultured material.
Activated charcoal can adsorb some inhibitory compounds, phenolic exudates and other metabolites.
Its use has been reported across a wide range of in vitro applications, including micropropagation, orchid seed germination, somatic embryogenesis and rooting.
Activated charcoal should therefore not be viewed simply as:
“the material that makes tissue culture medium black.”
Its presence can substantially alter the chemical environment of the culture medium.
3. Why Is More Activated Charcoal Not Necessarily Better?
This is one of the most important points when understanding activated charcoal in plant tissue culture.
Activated charcoal does not selectively adsorb only substances that we want to remove.
It may also adsorb useful components of the culture medium, including:
plant growth regulators, certain vitamins and some metal ions.
Earlier experimental work demonstrated adsorption of auxins and cytokinins from tissue culture media, while broader reviews have documented the complex effects of activated charcoal on medium composition.
Therefore:
Activated charcoal is not a “more is better” additive.
Whether it should be used, at what concentration, and at which culture stage depends on the plant species and the specific culture protocol.
A concentration that performs well in one system should not automatically be transferred to another.
4. Why Is Banana Added to Tissue Culture Media?
The second interesting detail from our visit was that the facility also used banana in the culture medium.
For someone unfamiliar with orchid tissue culture, adding banana to laboratory culture media may sound unusual.
In practice, banana is generally processed into banana homogenate or a similar preparation and incorporated into the medium as a natural organic supplement.
Banana can introduce a complex mixture of carbohydrates, minerals, vitamins and other naturally occurring organic compounds into the culture system.
For this reason, banana homogenate has been used as an organic supplement in some orchid tissue culture protocols.
5. Why Is Banana Associated Particularly with Orchid Tissue Culture?
Natural organic supplements have a long history of use in orchid tissue culture.
Depending on the culture system, materials such as banana, coconut water and other natural supplements may be incorporated into the medium.
One important characteristic of these additives is that:
they are chemically complex rather than single defined compounds.
Banana homogenate, for example, provides a mixture of naturally occurring components rather than one isolated plant growth regulator.
Its effects can therefore vary depending on the orchid species, culture stage, basal medium and other components of the protocol.
This is why it would be inaccurate to say:
“Adding banana always improves tissue culture growth.”
A better interpretation is:
Banana homogenate is one possible natural organic supplement used in certain plant tissue culture systems.
Its suitability needs to be evaluated within a specific protocol.
6. Is the Combination of Banana and Activated Charcoal Unusual?
Not necessarily.
The combined use of natural organic supplements and activated charcoal can be found in some orchid tissue culture systems.
What we observed during the facility visit was therefore not an entirely unusual or unknown technique.
For professionals who routinely work with orchid micropropagation, it may be relatively familiar.
However, this does not mean:
Orchid tissue culture = banana + activated charcoal.
Different orchid genera and species can respond differently.
Different culture stages may require different formulations.
Basal media, plant growth regulators and other additives also vary between laboratories and protocols.
Some systems may use both banana and activated charcoal.
Others may use only one of them.
Many tissue culture protocols use neither.
7. Why Do Tissue Culture Media Vary So Much Between Laboratories?
This was perhaps the most interesting lesson from what initially looked like a simple black culture medium.
Plant tissue culture is influenced by many interacting factors, including:
plant species
explant type
culture stage
basal medium
plant growth regulators
carbon sources and organic supplements
temperature
lighting
gas exchange
culture vessel
culture duration
and many other variables.
A medium that performs well for one plant cannot simply be assumed to work equally well for another.
Commercial micropropagation laboratories therefore often refine their protocols through repeated trials and production experience.
Over time, each operation may develop culture systems suited to its own plant materials and production requirements.
8. What Can a Black Culture Medium Tell Us?
At first, our question was very simple:
“Why is this culture medium black?”
The dark appearance led us to activated charcoal.
Speaking with the team revealed that banana was also incorporated into the culture system.
Looking deeper into the subject then revealed how differently these additives can be used across species and protocols.
This illustrates something important about plant tissue culture:
Two culture vessels may look similar from the outside while containing very different culture systems inside.
The vessel itself is only one component of the in vitro environment.
A complete tissue culture system is created by the interaction of:
Plant Material + Culture Medium + Vessel + Gas Exchange + Light + Temperature + Culture Management
Conclusion
A black tissue culture medium does not necessarily indicate an unusual or highly specialized technique.
When activated charcoal is incorporated into the medium, significant darkening is an expected visual effect.
The more important reason for using activated charcoal is its adsorption capacity. At the same time, that adsorption can also affect plant growth regulators and other medium components, which is why activated charcoal must be evaluated within the specific culture system rather than treated as a universally beneficial additive.
Banana homogenate is a different type of additive—a natural organic supplement that can be found in some orchid tissue culture protocols.
The key lesson is therefore not:
“Black medium = activated charcoal + banana.”
It is:
There is no single tissue culture medium formulation that is ideal for every plant.
Different species, culture stages and commercial production systems can require very different protocols.
For us, these differences are one of the most interesting things to observe when visiting real commercial plant tissue culture facilities.
